Since I'm playing with my kids and bought nearly all of the figures, we don't really have individual ownership that limits who plays with which figure. Therefore, our favorite way to play is to shuffle the character cards and then deal out an equal number to each person.
From there we have played a few different ways... when there are three of us, we play king of the hill with the winner staying. With more than three people we've just tried to do some sort of round robin.
One time we tried it Pokemon style, where we turned off the restore health option and then threw out our next Skylander against the previous winner. In that mode the new character pretty much always quickly wiped out the already hurt one, but I still liked the concept.

What a brilliant way to decide who plays who! Heh, my husband has both Terrafin and Stealth Elf (two of the best and quickest Skylanders after their HC are complete) and I have Drobot. Drobot is a great long-range character and can spam blades of rotating death to bounce around the arena, but he pales in comparison to the other two in PvP. It would be nice to kick his butt with his own character against his will when we aren't doing PvP trial runs to see which one preforms better.
Another way to randomize the characters to play -after the opponents are chosen- is to choose two and set them on the portal at the same time. When the character selection screen comes up, it is a surprise as to who gets which of the two Skylanders.
